Purchased three bedroom slab ranch home. House has natural gas furnace, stove and hot water tank. There is a dedicated laundry room that has only electrical outlets for washing machine and dryer. Want to run gas line from furnace/hot water tank gas line to laundry room. What materials will we need to accomplish this task? Also, can we use flexible tubing for the gas line? Thank you.

The vent flaps are easy to open (by design, it has to open with air pressure) so birds and rodents could easily open them up and make a home. Birds are relatively easy to evict, but rodents are likely to chew their way through the duct and find their way into other places. As for the crawl space vent, I wouldn't want a possible intake vent so close to to the dryer exhaust, it's a humidity issue waiting to happen. So because of that, I'd go for the roof vent. Try to clean it out yearly, and make sure to clean the lint trap out of the dryer between each usage.
The top hole is indeed used for the washer drain.
The lower pipe is used to drain a washing machine pan.
If the washer leaks this pipe allows the water to drain away safely, instead of flooding the laundry area.

My last house had gas plumbing where the homeowner ran a new line from the water heater line to the laundry room. It was red-tagged as unsafe.
I strongly recommend that you hire out this job; it's one of those "if you have to ask, you shouldn't be doing it" situations. Gas plumbing isn't one of those things that you can mess up and then fix later without any danger or problems.
