In this area there is a door with a lever and behind it seems to be some sort of arena.
During my normal first playthrough, this door was locked and the lever unfunctional.

I looked into this a little bit (youtube) and it appears that your understanding of the challenge is incorrect: you need to "not go into Last Stand Mode" rather than "not break Shield."
So you need a high level shield and a high high health value. I play as Salvador, and in my case, especially with some skills upgrade, when I activate the Gunzerker skill, my health bar recovers faster than I take damage.
So if you have a health regen skill, activate it right when the shield goes down, and this may delay your death.

To open this door first climb the nearby ladder.
Next look for a spot where you can jump the rail onto some pipes. Run down those pipes towards a somewhat hidden control room.
Finally, switch off the green levers to open all locked jail cells.

This arena is where Hyperius the Invincible stands around all day waiting for challengers. You will receieve a quest for this boss after beating all the story missions in the Scarlett DLC.
