According to the Prima guide you can win them in horse races.
In addition to saving the Northern Realms from the threat of the Wild Hunt, there are additional activities you can try; and two of these pastimes are fistfighting and horse racing. The former allows you to hone your unarmed fighting skills, and should be undertaken after reading up on the Training section dedicated to this activity. The latter enables you to equip Roach with additional saddlebags, saddles, and blinkers that help you store more equipment, increase your horse’s Stamina, and reduce Roach’s panic level. All are well worth attempting; the horse races as soon as possible.

Once you reach the bustling village of Midcopse, take some time to explore. They aren't meant to be killed. In the tutorial area they are there to be designed to teach you not to steal from people while guards are around. If they kill you you just wake up after a while and lose some money, you don't even have to reload a save.
If you kill them they simply "respawn" and go about their day.

After looking around at multiple forums and such, people have come to the conclusion that you just can't open the door. This has been deduced mostly by the fact that several people can't find it, as well as the fact that there are no windows of any sort to look into the building.
Traditionally (this isn't every case) when you can't see inside of a building in any way, there's no way inside.
Edit: Someone had said they got in when they picked the Master Key as their starting gift, but that has since been removed.
It's possible that this will be used in DLC some day and that's why the door is interactable, but as of right now no one has figured out how to get in.
