How much does armor diminish damage taken? Also, how is it's durability calculated? Is it worth making a set of diamond armor or is leather just as good (seeing as leather isn't used for anything else)?

Biomes are generated from the level seed stored in the level.dat file. This is one of the reasons you can visit maps from prior versions and find biome-like coloring (and why expanding the map will generate biome terrain).
The chunk terrain, however, is saved, so areas that were sand will still be sand regardless of what "biome" the game thinks it's in. Terrain already generated won't change, so if the level seed changes, you can find a grassy or snowy area next to a desert along the boundaries of the chunks.
First note that the Enchanting page on the wiki says that Protection, Fire Protection, Blast Protection, and Projectile Protection are all mutually exclusive, but it looks like you can have Feather Falling on boots with one of the other enchantments.

Since increasing the level of the enchantment only increases the EPF, when trying to find the "best" protection it is only worth looking at the highest level enchantment. The article also says that in order to maximize protection from a particular kind of damage, you would need 25 points of protection for that damage type.
Since there are 3 different types of damage, one would need 75 effective points of protection, but since each armor can add either up to 11 points to one type of protection or 5 points to each type (which is like adding 15 total), the max is 60 effective points of protection. So, you can't max out everything. And plain Protection can't be maxed out at all. Therefore, the best armor depends on what you are looking for:
If you want to maximize the minimum protection value:
Put Protection IV on each piece of armor. This will give you 20 points in every type of protection. As a side bonus, if you put any level of Feather Falling on the boots, you will max out fall damage protection.

If you want to maximize the average of protection values:

The way armor works in MineCraft has changed quite a bit since this question was last answered. This is an overview of how armor works in version 1.6.
A player has a total of 20 Defence Points (represented by the little "chestplate" icons above the health bar, where one point is half a chestplate). Each defence point reduces the amount of damage you take by 4%, but only for certain damage types. So, all 20 defence points filled up will reduce the amount of damage you take by 80%.
The type of armor (Diamond, Iron, Gold, or Leather) determines how many defence points are filled up, as well as the durability of the armor (how many hits it takes before the armor breaks).
You can also enchant armor, allowing it to absorb more damage, or even last longer before breaking.
