Everything I can find says that a Pokemon should become a Best Buddy once it has earned 300 hearts.
This Pokemon has 621 hearts and still isn't a Best Buddy. So what are the other requirements for it to become a Best Buddy?

The second menu on the buddy screen shows the number of activities done with your buddy, not the number of hearts. It counts every interaction you’ve done with your buddy, including the ones that don’t result in a heart.1
So while you have done 621 activities with your buddy, the number of hearts hasn’t reached the 300 heart requirement. Unfortunately (at the time of writing) there is no way to see the total number of hearts your buddy has acquired.
1 To easily test and confirm this, go to your buddy, check the number of snapshots, and then spam taking snapshots of your buddy. While you only received 1 (or 2) hearts, the number increased by the number of snapshots taken.
Compatibility with tablet devices is not guaranteed.
For whatever reason, they decided that your tablet does not "work" with the game for whatever reason, and have since blacklisted it from being used on those tablets (even though they meet/surpass the actual requirements for running the game).
I'd contact Niantic Support and ask them why your tablet is explicitly blacklisted. It may very well be a mistake on the part of the last published version of the app.
To receive the defender awards, you must have at least one Pokémon in an allied gym.
To collect the rewards for having a Pokémon at a gym, you have to go to the shop while at least one Pokémon of yours is still defending at an allied gym. Thus it is best to capture as many gyms as possible (up to 10) and then collect before any get taken down. To get to the shop, from the home screen tap the Pokéball icon, then tap "Shop". Then inside the shop, there is a button in the top right where you can collect rewards for having Pokémon in a gym. Note that you can only collect once per 21 hours.
Also, as pointed out by Virusbomb, "the rewards for defending a gym are 10 Pokecoins (which can also be bought with real money and are used to buy in game items) and 500 stardust. You can get a max of 100 coins and 5000 stardust every 21 hours with Pokemon in 10 different gyms."
