I have a page layout for a custom object. I have a related list on that page layout. The related list is open activities. Is it possible to sort the records in the related list by any column? Currently it is being sorted by the due date of the task. However, I have a new customer field called item number which I want to sort by.

Try to use the following formula of type "Text":
IF(ISBLANK(FieldToSortAfter__c), IMAGE('/s.gif',' ') , ' ' + TEXT(FieldToSortAfter__c))
The formula checks for the blank value and if it is then it puts a transparent image so nobody can see it on the page. But if the field has a value then just puts a space before it so the sorter "thinks" that this value is "smaller" that the image :)

It is not possible to display Related to field in the related list of opportunity page layout. Because, related to field is basically the opportunity lookup to which the task/event is linked. And in the related list, only those tasks/events are displayed which are linked( related to) to the opportunity. So, there is no need of the related to field. Let me know if this helps.

Workaround : Create a list button on Task
with URL value as /007?id={!Account.Id}&rlid=RelatedActivityList
Add the button to Open Activities Related list of Account Pagelayout.Then on Click of button it will open new page where you could sort open activities on any field.
